So I kinda messed something up :( I dehydrated my aphids with…..70% ethyl alcohol because I'm planning to mount it on a fructose solution(budget) and using glue to seal the slide making it "permanent". So wat do

Hi MicroMan,

Your post seems to have been truncated without including a description of exactly what your problem is so I will make a guess from what you have said.  If you placed "live?" aphids in 70% alcohol they will probably distorted and shrivelled due to their soft bodies and the osmotic forces between body water and alcohol strength, after distorting the insects the alcohol will have fixed them (precipitated and hardened their proteins) so they can never be returned to the shape they had in life, also any transparency they may have had will have been lost.  If you wish to mount aphids in a gum medium (water based) then I would suggest you try placing them (live) into alcohol no stronger than 30% and observe how they appear before mounting, if they look good mount them, if they are distorted try using a weaker alcohol solution.  If you wish to mount them in a resinous medium then kill them as above (30% alcohol) then transfer them up an alcohol series (50%, 70%, 85%, 95%, absolute) about a day in each, if distortion still occurs then you will have to either/and or fix them after killing, make the dehydrating series longer with smaller increments; once dehydrated the alcohol needs to be replaced with the solvent used in the mounting medium then they can be mounted.

Hope this helps.

Good luck

Peter.

Hi MicroMan,

Measure out 30ml of 70% alcohol then add 40ml water (to make a total of 70ml) and, wela, 30% alcohol.

Peter.
