why would a condenser need to be achromatic/apochromatic ? As I understand this is not necessary/helpfull. What is your specimen? ant head, it is reflective, because the light moves with the Z axis , the overexposed speckle on the ant facing the light also spreads... IMHO, this object is not transparent, so it is better photographed under top illumination, light coming from above. You might achieve it w...

is there nothing that can be done by changing lenses diameters and distances while keeping the aperture diameter and magnification? I tried light sheet, on 4 and 10x it isnt super usefull because the depth of field is enough for stacking software. On x40 (stopped down) I didnt get enough light throu...

Hello,
If you were given complete control over microscope (using dslr camera) and objective design except for the absolute aperture size of the objective, what would you change to increase the depth of field at the magnification given by a 40x objective ?

@hobbyist your results pictured above are consistent with mine. You had to push the iso (noise...) to see something, that's because most of the light is wasted. I suspect positive results were obtained using a much stronger illumination than mine in the connecting slide fixture, but i'm not even sur...

In order to get darkfield on x40 and x 100, I tried blasting light (just 3w led) on the edge of a slide a couple months ago, without much success. Even though I got great improvements by oiling the edge... In theory the change from air surface to water should help the light get out of the glass sli...

here are some last resort tips: -lower the iso if you're on a digital camera -make sure the iris, the stop, output of the condenser, objective are properly aligned (there are some tutorials and tips above) -make sure you dont have ambiant light parasiting ! -a good way to debug darkfield is to inser...

I havent succeeded stacking on regular 40x, I believe that's because the depth of focus is too thin for the software to process. I suggest you reduce the aperture of your objective to increase the depth of field
