Very nice pictures - looks like you used DIC or oblique lighting. I spent time down in Houston, but the water around Galveston wasn't very clearn I remember.

Hi - the best cameras for photomicrography are DSLRs, followed by C mount cameras and then cell phones. Hi the solvent used for amino acids was Deionized water and 95% Ethanol (Everclear from the Liquor store) in a ratio of 1:1 to 1:4. You need both water and ethanol. Other alcohols did not work for me. Only the callus remover needs to be spread out thinly, the amino acids will do this by themselves. ...

RE - Diatom colour shift - that is my fault. I shoot RAW files and when I process the images I must have used a slightly different setting, or I may have twisted the DIC compensator slightly between taking the pictures which changes the background colour.
