It worked, I made a mark on the diffused lens, aligned the condenser with that, the iris closure was not on the center when looking through the eyepiece, but when the eyepiece was removed and I looked through the tube I saw that the iris closure was in the middle aligned. I have the same a LED with a diffused lens on top, but I can remove it an see the LED (light bulb).
What if I made a temporary mark on the diffused lens, in the middle and I align the condenser with that, would that work?

How to check if condenser is aligned?
I was wondering these days if my condenser is not aligned, so I did some research on the internet and nothing. Nothing for light microscopy, so I ask you fellow members.
How to check/with what/ if the condenser is aligned?
Also I do not have a field diaphragm.

Great images, please John post the higher magnification photos, also you've done the second image just by varying the time of exposure to a stain or time of differentiation (and the second image is done with just safranin and fast green ?)

MicroMan2 I solved it, I mounted cartilage tissue in Nail Polish and it did not crack, nor it formed bubbles. Here's my protocol: After the staining, dehydrate in 70% IPA 1min, 90% IPA 1 min, 100% IPA 1 min then add the Nail Polish (I used one that had IPA as a solvent in it so it mixed perfectly) H...

I've been unsuccessful so far with Nail Polish, I tried to mount skeletal muscle stained with H&E but it cracked horribly after couple of days, or it made bubbles around, above and in the tissue when I tried to mount kidney samples. Best and most convenient is with Glycerol Jelly.

Well sadly I have no photos of that, but maybe my description will do. RBC were normally shaped (biconcave), although some were broken/shattered ( probably due to defrosting),and WBC none so far, but I only took 2 samples of the sediment and didn't see a sign of WBC, therefore I do not know if they ...

Crater Eddie nothing in particular, I just looked at the sediment that formed from centrifugation (epithelial cells, skeletal muscle cells and some erythrocytes but that seemed to be all, except this pollen sample)
