Re: "New" Zeiss, unsure of model

I see, indeed it is darkfield. At first I thought that was a phase ring because I'd never seen a darkfield setup before. Phase and fluorescence are pretty much all you see in biology research nowadays, otherwise just plain old H&E stains if it's medical doctors showing images. Well, hopefully I can ...

Re: "New" Zeiss, unsure of model

The condensers cat number is 465277, it appears to have phase 1/2/3, brightfield with variable aperture, and the rest are empty. Given the third phase ring, which I didn't notice before, I imagine there was at some point a compatible phase objective on here. Maybe I will go to where I got this and s...

"New" Zeiss, unsure of model

Haven't been here or used a microscope in years due to a change in my line of work but I've suddenly I've acquired this old Zeiss. I'm unsure of the model but it's definitely old because it was made in West Germany, not Germany. It has all air, 2x, 3.4x, 40x phase, 63x phase, the correct phase rings...

Re: Some Algae DNA

Yeah you don't buy 50mL of any kind of fluorescent stain. That amazon link has $40 shipping, and that's still oddly cheap. Thats probably for making a lot of DNA gels or some other application, maybe lots of qPCR. I'm not sure that you can just buy the stuff. It's typically 10000x concentration, i.e...

Some Algae DNA

I neglected to take white light photos, probably because this dye bleaches fast so I was focused on that. This was stained using SYBR green, we're at 40x. These cells occur in diploids. You can also see what I believe to be a nucleolus with some odd ringed structure around it. Not sure what that mig...

Re: Slide-surfing surprise - fungus! - maybe.....

I'm not too familiar with it. But at the very least, fixation and staining are chemistry based and the rates at which those things occur are subject to temperature. Typically shortened stain and fixation times reduce cellular deformations and increase penetration into paraffin sections. Usually I se...

Re: Culture medium for algae and other critters

Oh I wish I saw this sooner... My lab is an algae zoo, complete with various kinds of media (many of which are highly similar). Is this fresh or salt water we're talking? There's lots of recipes out there, but you kind of need a small chemistry lab to make them. You can probably boil solid fertilize...

Re: Slide-surfing surprise - fungus! - maybe.....

If you look at root parts you should be able to find fungus pretty easily, pretty much all plants have symbiotic fungi. Leaves, probably not so much. Have you ever considered microwaving your samples while staining or the stain itself? I know people do that for EM and sometimes with giemsa for regul...

Re: looking for a better camera

I think using SLR's is a good choice. Mirrorless cameras would be better probably, I know someone who uses a Sony Alpha as a microscope camera it's it's really nice. For bright light, I think regular photographic cameras are better than dedicated microscope cameras.

Beautiful images, kinase. Is the last image a 'stack' of images...or did all the tendrils just happen to lie in near same focal plane? Where was this algae sampled from (the adjacent areas seem so clear of debris)? For your images here..what brand/ what spec coverslips do you use? thanks for these p...

These are Guillardia theta which is entirely unrelated to Giardia lamblia. That's easy to tell if you look at the names, but hearing people pronounce the genus might lead you to believe otherwise. These have phycoerythrin. Hence when I hit them with a blue light, they fluoresce in this nice yellowis...

Re: AmScope T690C-PCT200-PL sufficient for spirochaete viewing?

Investing a fluorescence setup just to do that is a huge waste of money, especially if it's not going to be Zeiss, Leica, Olympus, or Nikon. Where would you do this to keep it contamination free? Have you done fluoro imaging before? I don't normally use fluoro stains to look for bacteria, but I can'...

Re: Oiling the condenser to the slide

I'm not quite so sure how this is all going to work. The olympus rep said we could certainly do it with the IX73 we have but I think the PI is going to opt for trying to get the condenser for the Zeiss because that'll be way cheaper in the short run. I know one person who bought a high NA condenser ...

Re: Oiling the condenser to the slide

Actually we got in contact with someone who has a 1.4NA condenser, even better than what I was hoping for. It's an old Zeiss inverted microscope, I think an Axiovert 100M. I have a 40x and 100x of NA~ 1.4 for it so that condenser should work great as long as it still has DIC on it. The current .5 on...

I'm having this issue. I have an oil 40x and 100x both with an NA of around 1.4 and the condensers NA is like .5. I need to see If we can get a better condenser. It's hard to find info about condensers on the manufacturers site, anybody know where they sell one? It's for very new scopes. I think I m...

When you guys have issues, is it when trying to take photos of stuff that's moving or stuff that's still? Unless I'm doing cancer stuff, which means it's probably fluorescence and fixed and embedded in a hard setting mountant with DAPI, then it's usually stuff that's moving. I found some odd corkscr...

I even have this problem and I'm using this scope and this camera. I pretty much only use phase, DIC, or fluorescence. Now that I read I can see that it's not truly a 17mp camera but using pixel shifting to achieve a higher resolution. I'm truly not sure what causes it, I've tried as much optimizing...

Re: Quick test of new micrometer slide

The confocal I use can put the scale bar an any other measurement you desire on the image. Usually in papers, it's just a bar that is out of the way of the subject in a corner, usually the inverse color of the background. i.e. brightfield usually has black and fluorescence with black background whit...

That'd be my initials carved (not sure that's the right word) into the nucleus of a single cell. DNA is blue, a protein that localizes to sites of DNA damage caused by the laser. The laser microdissection microscope is even more precise than I thought. I actually tried this a bunch of times in a bun...

Re: what is this

Probably just stuff moving due to Brownian motion, either it was on the slide before you put the sample on, on the coverslip, or if you added water it'd have been in there or from somewhere else. It'd be pretty hard to keep out absolutely everything from getting on your slide in your house.
