Did you mean 40 W tungsten or tungsten-halogen? If so, I would be concerned about heating your objective too much. Did you try first with lower power objectives? The 40x would be so close to the object that it leaves little room for direct light from the lamp; lower powers are easier and would be w...

I'd like to use the GUF but my filter holder only accepts a round filter whereas all I see are rectangular strips. I can use Photoshop CC to create a round gradient but I don't know whether that would work or not. Any suggestions?

Thanks for the heads up Phil. I just thought it would be useless to slice right in to the cell or by slicing, capture two cells on top of each other or say slice the top off a cell if it's standing vertically. I've never done this before so I guess I'll know soon enough as I practice.

You could try this. Your diaphragm has a series of hole in it instead of an iris with metal leaves that can make one hole larger or smaller.
