Re: Can't see sperm in microscope
Well 40 years ago when my wife and I wanted kids and couldn't conceive, I, too went to the microscope. Same results as you! We went to the professionals and they too took sperm samples and examined them. I could have saved a lot of money, because the reason I couldn't see my sperm is because it wasn...

Re: Incident oblique light on a specimen
I'm ahead of you there 75! I'v made some stops out of expired gift cards and have used them up to 40X with a great deal of success. I have one in my Infinity Plan MS right now. Compared what I got with an online image from MicroscopyU's image of a Phase Contrast image of epithelial cells and m...

Re: Incident oblique light on a specimen
Thanks, Fan. rather than throw money at what might not work anyway, I think I'll forget it and go back to oblique illumination along with Rheinberg Illumination.

Re: Incident oblique light on a specimen
Did you mean 40 W tungsten or tungsten-halogen? If so, I would be concerned about heating your objective too much. Did you try first with lower power objectives? The 40x would be so close to the object that it leaves little room for direct light from the lamp; lower powers are easier and would be w...

Incident oblique light on a specimen
I love to experiment and when I saw this article on Micscape UK site I had to try it: http://www.microscopy-uk.org.uk/mag/indexmag.html?http://www.microscopy-uk.org.uk/mag/artsep09/rv-darkfield.html It's really simple but I can't get it to work. He uses a 4 watt LED on a gooseneck to get close to th...

Re: OMAX 5MP A3550U Camera & ToupView software
Yep closing the apps helped. Also I noticed when I use my DIY/DIC stops, there's less light and its harder for the camera to focus. I have to use the condenser iris full open in some cases and rely on the stops to get contrast. In the photo above, I dont know what causes the purple color, its not pr...

Re: OMAX 5MP A3550U Camera & ToupView software
Well I tried changing to a lower res for the view and a higher res for the snap but It's actually harder to focus with the lower res. I think as, rabitt says, Processor speed might be the problem. In my case my processor is running close to 2.67 MH and I am running Win8( I refuse to upgrade to an in...

Re: OMAX 5MP A3550U Camera & ToupView software
That's great Rich, you'll soon be up & running - Toupview is superb. :) I wish I could say that John :( I now have three Amscope cameras that run Toupview. I'll admit that the software does have a lot of features but I can't seem to focus it so the images are sharp and it definitely is not parfocal...

Re: Phase Contrast on new scope
Some phase is just not very good and some to the low end Chinese stuff is even below that. I It's not that. I didn't just buy the "Chinese stuff" because I couldn't afford anything better. I checked on google images searching for "epithelial cells Phase Contrast". My images looked as good as those ...

Re: Phase Contrast on new scope
Thanks 75. I am in contact with Amscope and they say they will give me a partial refund if I send the Phase kit back. The T690 scope that the PC kit is bundled with it is a better scope so I'm just going to keep it. Amscope is very strict that the scope must be packed up exactly as it was shipped an...

Sorry fellas  I was taking a hiatus while my other half had things for me to do, Just got around to reading(and laughing) all the suggestions. Phase Contrast on new scope
Hi I have just received an Amscope T610 Infinity Plan Microscope bundled with their Turret Phase Contrast kit. I am having trouble setting up the annulae for Phase Contrast. I replaced all the objectives that came on the scope with their Phase equivalents. I rotated the condenser wheel until each ri...

Re: van Egmond Mask Experiments
Oh I see! I was calling it DIY DIC and Oblique illumination! I took a closer look at that first picture and I have made that same patch. I use it the most often. Was going to post some pictures when I get good enough at it. I just put mine in the round filter holder but I can't adjust it that way. I...

Rod I can find nothing on the web, other than your videos on Youtube. from your pics here they look like a pattern of variable spaced dots sort of like a magnified picture of a newspaper photo, could you posts some links about them? Where did you first hear of them?

Re: van Egmond Mask Experiments
Rod I am fascinated by the results obtained by the use of simple patches and filters. I got some black cardboard and am experimenting with cutting out different shaped openings in my patches. Just looking at cheek cells and seeing the actual contours of the cells is exciting for me! I am intrigued b...

Re: GUF #2
Thanks, I;ll try those suggestions. Since I've tried making and using various oblique filters I'm constantly replacing filters and I have to unmount the condenser each time, I'm thinking of using a nikon condenser that uses a slider so I can quickly change filters. My condenser takes a 37 mm condens...

GUF #2
I'd like to use the GUF but my filter holder only accepts a round filter whereas all I see are rectangular strips. I can use Photoshop CC to create a round gradient but I don't know whether that would work or not. Any suggestions?
Thanks,
Paul

Re: best wattage of lamp used for phase contrast ; Thickness of a plant cell
Thanks Charlie! It will probably be a coupla weeks for both, but certainly not longer than that. I'm like a kid at Christmas! Mean while I'm going to the hardware store and get a large nut and bolt and a straight razor and practice with that. You can get a straight razor with a replaceable blade at ...

Re: best wattage of lamp used for phase contrast ; Thickness of a plant cell
Thanks for the heads up Phil. I just thought it would be useless to slice right in to the cell or by slicing, capture two cells on top of each other or say slice the top off a cell if it's standing vertically. I've never done this before so I guess I'll know soon enough as I practice.

Re: best wattage of lamp used for phase contrast ; Thickness of a plant cell
Plant cells are delicious. Red or Golden? Thanks Lorez! Well I was looking at Amscope T690 Which is Infinity Plan and comes with a PC kit designed for the scope. It has Full Kohler and I like my present T400b just fine so I think I'll like this one also. I finally found out the thickness of t...

best wattage of lamp used for phase contrast ; Thickness of a plant cell
I'm going to sneak two questions in here for the price of one! [1] I'm purchasing a new microscope with phase contrast and infinity plan objectives. The scope I have now has a thirty watt halogen lamp and seems bright enough for brightfield viewing but this new scope I will be getting has only a 20 ...

Re: camera newbie questions
Well They all have a diaphragm of some sort! With my scope I get changes in contrast by opening and closing the iris. While I am sure the monocular scope your son has is a good scope(most Amscopes are) To get some real benefits you need a more expensive scope. I would suggest one with an abbe conden...

hey Great idea Charlie! I think the quality of the light source might be better, if i can find one. I think a microscope with a built-in LED might be in my future. What do you think of this one? It has Phase contrast with all the annular condensers built in, Infinity Plan also. http://www.amazon.com...

Hi! I just wanted to post a pic of my latest purchase, an LED light kit! I got it at the dollar store. Installation is simple. Re: camera newbie questions
You could try this. Your diaphragm has a series of hole in it instead of an iris with metal leaves that can make one hole larger or smaller. I don't know how many holes your diaphragm has, but put it near the middle hole. focus the scope on your specimen. Now move the diaphragm to each smaller hole ...

This thread is very interesting! I don't have a hand microtome yet but I want one like the one like you have. I have gone beyond onion and want to see the cells of other plants. Do you mind if I borrow your thread for a little while and ask a couple of questions? If I just put a plant stem in length...
