Yes you're right, edge's sharpness is also important for stitching. I am thinking that i can crop the best clear area from the bigger images (from the vignetted) maybe this work but I am not sure without try the camera I think Xline Olympus objective is good for this kind of situation ( but very exp...

Mu purpose is seeing bigger field of view. For example when scanning 15mmx15mm area, I have to take 650 photos (0.75mm x 0.55 mm). If I see bigger area for example (like 0.85mm x 0.75mm) I need less image and as a result less time to scan a tissue

@Hobbyst46 actually it is a scanner, objectives's FN value is 26.5 Tube lens information: Focal length : 180mm Projection area: 26.5mm, Focal Plane Position: 151.3mm 0.74mm obtained from 40x 0.95NA objective. First of all thank you for fast reply

Our equipment consist of ;

40x UPlanApo 0.95NA objective is purchased from Chinese
0.35X C-mount Camera adaptor and 1X Tube lens (both of them are from Olympus)

Actually the image on the tube lens is bigger than 0.75 x 0.55 mm, but camera has limited f.o.v.
