Re: DIY epi-DIC

Well done, Wes. It seems to me that it might be able to prepare many biological specimens for epi DIC as well. Slice 'em very thin, stablize without a cover slip, maybe even a combination of reflected and transmitted light? Thanks Pete. Maybe teeth, bones and shells would light up well with this te...

Re: diatoms

As I understand it you want a good difference between the refractive index of the sample (diatoms in your case) and the surrounding medium. These differences cause a phase shift (invisible to human eye) between the light fronts passing through the sample and the ones surrounding it. The phase shift ...

Re: Diploneis bombus

Thanks for the advice. So isopropanol does not affect the cement of the front lens, thats good to know. A long time ago I ruined a regular old 100/1,25 achromat using ethanol and I've been ever since paranoid about solvent choice. EDIT: so newer lenses from Zeiss (whats currently in sold by them) ar...

Best case scenario would be to retire early and set up a dream lab in the basement. Like these fine gentlemen have done (check out t...

DIY epi-DIC

I had the idea to build an epi-DIC setup with components I currently have namely a IIIRS fluorescent condenser, a couple of polarizers, a 50:50 mirror, a 16/0,32 epi-Plan objective and a 16/0,32 Plan DIC slider for regular transmitted DIC. In epi-DIC a single DIC prism is used to shear and then reco...

Getting a good shot of the nucleus in Spirogyra is no easy task.

What microscope and camera have been used to generate these?

Re: Two ciliates from my plancton aquarium in phase contrast

I'd really like to get a 40/0.9 multi-immersion Plan-Neofluar Ph3. These are some of the finest lenses ever made. The 63/0.9 is also on my list.

Unrelated I'd also love to get a Jamin-Lebedeff kit but I guess I'll have to wait a very long time for that

Re: Two ciliates from my plancton aquarium in phase contrast

Hi Bob,

40/0,75 Neofluar Ph2 is the only phase contrast objective I have and I too like it a lot. Its great for things like bacteria and algae but also protists especially amoeba.

Re: Assorted algae

Thanks Hobbyst! Yes, this is done with phase contrast (taken with a 40/0,75 Neofluar Ph2).

Its a fortuitous assembly of algae, the one and only occasion where I saw such a thing. I'm a bit puzzled as to how it happened.

Peritrichida

Went to my favorite spot a couple of days ago to collect water samples and found these peritrichs to be very abundant. They formed very large colonies on the side and bottom of the sample containers. Not sure about the species. https://kopame.com/dmqk/dmqm/media/peritrich4780medium.png https://kopam...

Re: Fluor vs Plan Fluor?

The correction collar on the Fluor objective is probably there because of the high N.A. The Plan-Fluor has a lower N.A. and therefore doesn't need such stringent control over coverslip thickness. If you plan on doing photomicrography I'd go with the Plan lens. If you just look for eye candy on the g...

Stunning! Is there a special name for the triangular structures visible in these photos and how did you produce the colors in the second image? This is all very, very interesting, a bit more detailed info on your epiDIC setup would be greatly appreciated as I'd like to invest in one.
