Aeolosoma

16x Objective, Oblique illumination. Filmed this, this morning. There are a few specs of dust "somewhere". Tried to find and remove them twice without any luck. In the end decided to continue filming, not wanting to miss the first Aeolosoma I had seen in a while. Love their bright colours. Video is ...

Re: Spirogyra

What are the grey steaks and blots on the second image? Not exactly sure what you are referring to. Photoshop does sometime outline "areas" when it blends images. If you open the second image in Photoshop and use the Magic Wand on the background they will show up. These areas are not normally visib...

Re: More Spirogyra

Lovely!! (but I think the width is actually 43.917 µm :) ). I plead guilty on all counts to getting carried away with my Reticle ;) In my defense I could point out that each division on the 25x objective is 4µm, if the alga is 11 divisions wide what is a poor Reticle user to do? "Why, round up or d...

Re: Culturing Algae

Amazing setup. I was expecting a petri dish! For now just a couple of questions. How many pediastrum do you need in a sample to start a culture from pond water? Are you careful to remove other algae from the sample before you start? Lastly, you seem to have more pediastrum than one would need to obt...

Thanks gekko and vasselle, will look into it.
I suspect the top polarizing filter (the one above the objective) will be the challenge.

Re: Spirogyra and Trachelomonas

Thanks vasselle and gekko. gekko, that link went right over my head :) Managed to dig out the phrase "cytoplasmic streaming". I take it that could account for the movement? Perhaps I should have tried additional techniques, problem is so far I only have BF, DF and Offset DF (Oblique). Hope to add Ph...

Spirogyra and Trachelomonas

16x objective, alga width 50µm, Trachelomonas 20µm, Darkfield.
Apart from what some might construe as a seasonal image – reddish, green and twinkly things – anyone know what the moving specs within the alga envelope are?

Interested :) Thanks

Re: New project: LED conversion for LOMO Biolam

Hi microgreen. A few of questions. 1) Are you dimming the mains current before it reaches the 220v to 12v ac transformer? 2) What is the DC source? How many volts/amps. 3) Details of the led bulb? 4) Have you taken video with the led light? Any Pulse problems? 5) Are the items I circled in yellow, f...

Hi Dale. Still toying with the trinocular option as I have a lot of camera equipment and experience. I was bitten by the microscopy bug in May. Have therefore been sharing my binocular head with a camera on a tripod for several months now. I strongly urge you get a trinocular head if it is within yo...
