Re: Hoffman Modulation Contrast
Hoffman avoids the costly prisms of DIC, that is the main bonus. The book, Light and Video Microscopy by Randy Wayne has a great chapter on it. It only uses a polarizing filter, retardation film, and slits in filters to to convert phase differences in the specimen into visible differences. A slit fi...

Well, I know nothing about said Biddy, but this is a the fine tome that I use to unpick an understand the binomial names in my Botanical pursuits - I find it very useful indeed to understand the meanings that are so often considered prosaic although personally I find a certain charm 'in there' also...

Duck weed needs lots of nitrogen. I use some limestone or clam shells to help buffer the water's pH, and add either soil or a tiny amount of fertilizer. I good general book that I like is: A Sourcebook for the Biological Sciences. https://www.amazon.com/Sourcebook-Biological-Sciences-Evelyn-Morholt/...

That is a species of Penicillium. Personally, I prefer fuchsine in lactic acid. No phenol, the lactic acid is a fixative and thick enough to take days to dry out, and has a different RI than the stained fungal tissues further enhancing contrast.

Stunning work! from what I scantily read about mosses, I thought that they do not possess conducting tubes... As for the images, they very nicely and clearly convey the botanical information! I notice that the leaves of your moss sample are quite large - 5mm long or so? BTW: Your "expensive" forcep...

Good to know more brands, but using "review" in the title is a bit confusing. I had expected more than an overview of the companies.

Re: Sporangium of fungi?
It is a member of the family Mucoraceae, likely a Mucor , Rhizomucor , or Rhizopus . It is an immature sporangium though, notice how thick the wall is and the lack of obvious spores inside. You need the entire structure, intact, including the point of origin on the hypha and preferably, a mature spe...

Sounds good. I used a tiny 1.3 mm drill bit and a hand push drill to drill out the majority of the piece. A trade sounds good. Just let me know what parts you want to swap. You might need to re-tap the hole for the knob in the condenser. I tried to be careful, but I probably brushed up against it wi...

I have a canon 60d and an ortholux with a trinocular head. I use this:

It works in any eyepiece so I do not need all the other attachments. It is a built in 2x lens.

Ah, I see. I was curious if you had one of the special rigs that move the camera a known distance. I have yet to try just putting my camera in the port and turning the fine focus for stacking. Good to know that you can do that and get quality images.

What do you use to control the focusing while taking the images for stacking? Do you use a normal canon lens for photography or do you use a specialty adapter lens, such as this: https://www.amazon.com/OMAX-Microscope-Adapter-Canon-D-SLR/dp/B01D914LT2/ref=sr_1_3?ie=UTF8&qid=1539802448&sr=8-3&keyword...

Re: Questions about Wild Heerbrugg M11 microscope eyepieces and light source
Hi, I actually am a mycologist. I would strongly recommend buying a objective that is around 50x to 70x. On a Leica DM2500 (if I recall correctly) I was most found of the 63x objective. It provided a much better image of spores than the 40x and still did not need oil. You can use reticles in Huygens...

Re: identify a microscope DIC or Phase Contrast
You can get results that give a 3 dimensional appearance ( similar to DIC) to the specimen by using oblique techniques. oblique techniques can get good nice picture? vs phase contrast which is batter? Depends. I do like oblique and phase. Still DIC will give better results for technical analysis, b...

Ortholux tube lens (1.25x)
I have been trying to find out exactly what a lens in my ortholux is doing. It is a 1.25x lens that is in between the objectives and eye pieces. This is an old black enamel ortholux with 170 objectives. Is this the lens people refer to when talking about creating infinity correction when using non-i...

Re: disassembly of Carl Zeiss Jena Amplival phase contrast condenser
Some advice was provided at the workshop that is different from that some of that offered on this and other fora. E.g., optical components such as objectives are cleaned primarily with 70% isopropyl alcohol and Q-tips cotton swabs (that brand only). That is in line with the Zeiss in house cleaning ...

I think we really need to know how you are attaching the camera first. I have used a Canon 60D dSLR and had no issues with dark field like this. I used an adapter that attaches to your camera like a normal lens and fits into eye pieces or C-mounts. $60 on amazon. Something like this, but I bought it...
