In my experience, closing the field iris a bit and using bigger patch stop disks yields a more DOF in the final image. Adjusting condenser height also helps better contrast. Crisp and well illuminated. Nice video KD.

I suggest try different size dark field patch stop disks for optimum depth of field and background darkness. I guess the last microorganism in the video is a Paramecium.

It's a very nice looking beast. You attached two analogue/film camera at both side of the microscope. Do I see and recognize them correctly? What's their purpose when you have a another digital camera on top of the ocular?

The objective lens you linked in the first post has the NA 0.65 which is in the range of achromatic objectives. I though apochromatic objectives have higher NA. Is my assumption correct?

A good quality 10 Watt led (e.g. Cree) will suffice for most dark field, DIC and phase-contrast situations when taking pictures. For observation only even 10 Watt is overkill in most configurations. It appears as if your stains have mixed and formed a new colour instead of attatching to different structures.
