I reworked the image from the TIFF-files and cleaned up the background and some edges in Lightroom. I do not think it ended up any better. Each TIFF is 100 MB and the JPGs are like 4 MB. Pictures would help. Usually there are plastic or wooden blocks on which you mount the wax. Then you clamp the block to the microtome. At least that is how mine works.

The safest way to clean diatoms is to call Klaus Kemp and ask for a ready made mount. Not as fun though as DIY. (joking of course)
