Re: Unidentified

Well zzffnn and gekko, it is a wet mount, also I added more water to the slide and nothing ( so not a crystal of m.b.). It appears to be stucked to the slide ( I fixed the cells with a lighter). The thing is the slide was pretty normal ( muscles cells, epithelial cells) and all of a sudden this. I v...

Unidentified

Hi there I just found this while centrifuging a blood sample from a beef. It is stained with methylene blue and is around 35 microns. Does anyone have an idea of what it could be?

Safranin working solution?

Hi guys, I recently bought Safranin as a powder, and I would like some advice on how to make the solution (if aqueous or alcoholic) what proportions should it have and what concentration of the dye. I am looking to do Gram stain but also general nuclear stains and cartilage stains. Please if you can...

Hi charlie g thank you very much, indeed I learned a valuable lesson (not to say: o, I will identify this organ after the staining :D). Thank you for sharing that image, it was kinda the same that I saw and decided that the chicken will serve a even higher purpose for science (also cooking). Nonethe...

Re: Unknown organ identification, H&E stain

I believe that as charlie g named it so poetic, Viscera unknown is a lymph node. I searched thorough a lot of histology book and all indicate this. It was hard to distinguish because my section was not sectioned at the proper size of a 6-10 μ, instead with my hand made microtome I think it is around...

Unknown organ identification, H&E stain

I've been having a go at making my own histology slide, but I encountered an interesting problem. First, the organ tissue was taken from a frozen whole chicken bought from the supermarket, it still had both lungs, both kidneys and what I tought to be a blood vessel from one kidney, I will post pictur...

Fantastic pictures, book quality there :) I'm glad to see you're still excited by your work. Keep up the good work.

p.s. the methylene blue pictures seems to have a little metachromatic effect on it, do you know a method of emphasizing the effect?

Raul.

Well it kinda looks to me like it, having dendrites, but the astrocytes are only present in the brain and spinal cord, it dependes from where this sample was taken.

Eosin proper concentration for histology?

As the title says I want to know what the suitable concentration of Eosin would be for most histological applications. I checked on the internet and some proposed 1% other 0.5% and even 0.2% (of course the staining time differed with each concentration). I would wish to know from someone that had go...

What was it stained with? Also can you take a picture of an astrocyte (I think I can see one about 2/3 to the right-down corner). It is a very nice sample of tissue, a beautiful piece and a lot of work and trails. Please post more :).

Thank you for showing this.

Recipe for a Romanowsky stain solution?

Hi guys I want to make the solution using methylene blue and eosin. I am interested in the proportions to use and also other aspects like buffers or oxidants for m.b.. I know that I can buy a cheap Giemsa stain, but I want to explore the science behind it.

Thank you.

Well done. :) Thank you very much, also you are a bit to blame John B :) that I bought H and E (giving me links to some sites, and more other advice)....

Thank you very much for your kind words, mrsonchus and JimT, the magnification was 400x microscope and 2.5x from camera. I have another sample with a bigger camera zoom 3x( this time on purposely overstained with Mayer's Haematoxylin) to have a better visibility of the nuclei. Although the striations...
