Excellent images - they really look spiny! Great preparation and EDOF. They show that good stacking - good in terms of giving information about the subject - isn't always a matter of throwing in as many images as possible, but a wise choice of constituent images - and you've definitely got that skil...

Hi John, Thanks for going to all of the trouble of providing pics of your setup... I didn't mean to cause you that much trouble... I think an outfit like that is a little out of my price range at this time... It is a major production for me to take pictures through the ocular.. I have somewhat lost...

Re: Lux meter

Hi John, I have to admit you get great photos with your old Kodak... For some reason I can't use mine as well.. It usually takes at least 10 tries to get anything usable.... I think I have become somewhat shaky in my old age... What is your digicam that fits into the trinoc?... Ah bill, it'll just ...

Hi John, I am sure you are on the right track with your camera setup... You probably have a nice DLSR to use... I have a point and shoot cheap very basic camera: It is for very informal photography... Hi bill - nooooo.... no DSLR here! I had a beautiful Canon 1200D attached to my trinocular main 's...

Re: Lux meter

I usually compare (or attempt to compare :) ) light-levels with my camera set to auto-exposure with a 'target exposure' set (in software) and kept constant, the camera (in my case a 2mp eyepiece UCMOS USB) then sets the exposure time accordingly, this varies as expected when for example I change pow...

Extremely interesting post - thanks for posting it. I'm going to have a good look at this subject as soon as I get time - it's obviously very important to get the priorities of all the various factors clear and to attempt to optimize them as much as possible with one's available kit.

Hi Manfred - those are good images - I like them a lot. :) They're reminiscent of the look of tissue-sections whilst they're still encased within wax. I've tried the nail-polish impressions myself, but a long time ago - your fine pictures have made me want to have another go!

Thanks everyone! A stage micrometer looks like a great investment, I sometimes try to calculate the size of organism but it is tedious. PVDF translates to Polyvinylidene Fluoride The uses and varieties of both Polyvinylidene Fluoride and Nitrocellulose membranes are somewhat elucidated (and priced) at Bio Rad Laboratory's website advertising them: http://www.bio-rad.com/en-us/category/western-blotting-membranes-filter-paper?WT.srch...

Hi, very interesting technique, and so simple in principle, given the appropriate membrane and reagents. I don't suppose, considering the charge-barrier that you've encountered with the membrane, that you could apply a 'negative-staining' (negative in the optical sense, not the charge) technique to ...

Hi John, Walter's book is on it's way to me. https://en.wikipedia.org/wiki/Monochromator Your images are very good indeed. However, regardless of tethering, parfocality (between eyepieces and camera sensor) is essential in order to preserve the optical corrections of the microscope optics. Only then will you you be using the objective as desi...
