Thank you all for your kind words. I cut approximately 2-3mm off the tip of root. Then I placed the root tip on a slide with 1-2 drops of 10% Hcl (muriatic acid) for 5min. After 5min I added 1-2 drops of Hematoxylin for 5min. Then I removed Hcl and Hematoxylin with paper towel and added just one dro...

It looks like DIC,you can get different colors by moving the upper DIC prism.75RR wrote:Did you use an additional filter or can DIC produce such colours?

You have a good eye,I had made a mistake in the calibration software.
First image was little polarized light,rest DIC.

I see that you do not surrender, that's good. In my opinion the sample pieces were too big and did not manage fixing and dehydrating. My sample pieces were not larger than 3-4mm. After fixation and dehydration through a series of alkochol the sample lose their chlorophyll and become almost transpare...
