Found these guys floating around while slide surfing lastnight. Both images are from the AO20, oblique illumination with planapo 40x . 80 / adv planachro 100x . 1.25 I was short on time so i didn't oil the condenser or 100x objective. No doubt it would have improved image quality. I'm in the process...

Rodney - You bring up a very good point. There is little room for error when using fast drying super glues. You have to be spot on and confident you have everything lined up and in the right position before you commit to placing the glue down. Thanks for the suggestion of a slower drying adhesive! C...

The grey tube is just slightly larger than the opening on the t-mount for the EOS adapter....Industrial strength super glue was my friend during this setup. Following some advice over on the "camera vs eyepiece" discussion i've managed to finally get my camera/eyepieces parfocal and almost the same FOV. I think the image quality i'm getting via my camera has improved because of this. I have to crop a small amount but overall i'm happy with the results. ...

AO20, 40x .80 Planapo, Oblique Illumination, 3 panel composite...each panel is a 5 image stack, Stacked in PS. Panel blending and stacking got a bit wobbly in some places but it's getting late and i'll end up making a bigger mess if i start playing around with it!

zzffnn - It could very well be that the filter was not entirely parallel. This is all experimentation right now it's something I'll look at. Oh neat! I like what you've done with the pvc pipes! You could try making the pipes longer so that the filter is right below your stage and then try different size black spots for the dark field. That might give better results for the higher objectives.

Your are right, the pollen does have a 3D effect to it! You got some great detail, is it a single image or stack?

Thanks for posting the original of the xylem element cell-wall, its awesome to see the additional detail that post-processing can bring out!

It's a really great collection of images! Love the difference colour can make. I especially like the black and white one! It's shows great contrast.
