A biology dissection kit has some nice tools. There are a few of them on Amazon. And not too pricey. I got this one. Practising microscopy techniques on sauerkraut bacteria.

Should posts like these be posted in the sandbox? Practising microscopy techniques on bacteria. Heat fixing, oil immersion, methylene blue stain (staining). Image one seems like I was looking at yeast cells. Over time the bacteria seem to have gotten smaller? And seems like I could focus for a bette...

Thank you I was up most of the night finding distinct critters. Look like I also found a leech or two. I am new to microscopy and have a very limited vocabulary. Is Oblique the same as phase contrast? Does it require another gadget? I have phase contrast but images do not seem to appear as sharp -- ...

What are some of the most popular stains (please include wet mount media also)used in Microscopy?

I am taking a poll on the most commonly used stains in microscopy. I'll summarize after a few posts. Popular Science has a similar article -- that might need to be revisited. Here is what I have on my list including wet mount media: 1) Bismarck Brown Y 2) Eosin Y 3) India Ink 4) Iodine Solution 5) J...

Thank you Hobbyst46 and mrsonchus for your feedback. I appreciate tips!!! The Bismarck Brown stain looks suspiciously rather cellular. I need practice looking at and dissecting stuff.

Thank you for your time and explanation Hobbyst46!! I have a lot of fungus and a few more stains. I'll do a visual comparison of various stains at time t0 and tn and see what happens. It took a while until I noticed the behaviour of the iodine. I'll see how other stains fair. Something wetter than w...

I am assuming that there must be some sort of nifty esoteric explanation. :geek: Like what is being absorbed? The fungus is growing on carrots that were stored in a plastic bag. I kept the fungus growing for further investigation and to attempt a positive Id of the fungus. Sclerotia is a bad omen fo...

Any ideas of what is going on with fungus and iodine?

Just wondering about what is happening. I place a drop of Iodine on a fungus (I think sclerotia) and the iodine drop appears to be clearing and the fungus is absorbing a colour. Is this some sort of a cation exchange? The iodine drop beads but something is being pulled out of the iodine. Tried a few...

A Large Something Or Other In The Vermicompost Tea!

A critter in vermicompost tea after 8 days of aeration. Is it a Red Wiggler earthworm (Eisenia fetida) or a Nematode? From previous observations when I aerated I saw that Red Wigglers die off and float to the top then sink. I do not think a red wiggler can survive in compost tea. So do not think it ...

Re: Vermicompost tea after 8 days of aeration.

Even at 40x with no well slide, the organisms are just shadows shooting around.
What kind to the medium can they be mounted in that will dry with no air bubbles?
Would something like glycerol work?
Or just let them dry on a slide without a coverslip then apply coverslip after drop dries?

Vermicompost tea after 8 days of aeration.

At 20X, a 40X Objective bends the coverslip on a concave slide.

Looks like a nematode survived 8 days of aeration at 15° to 20° c temperature.

Very small critters in there!

I have found a way of estimating nematode populations, stain with iodine, let them die. take picture(s) and count on picture them. Graph paper (squared paper) as background can help.
