Re: Amoeba (video)
Many thanks, 75RR. On my E-P1, what I do is set "custom white balance" in still mode (I use Aperture Priority because I don't use an attached camera lens) using a blank (clear) area on the slide. I'm not 100% sure, but I've been assuming that the white balance set in still mode also applies to video...

Very nice. What image editing software do you use? I ask because, at least to my taste, your image, excellent as it is, can, nevertheless, be a bit more contrasty and sharpened slightly.

Re: Help with identification
75RR, thank you for your suggestion. At first blush, I also thought that the protrusions (including the "stems") were pseudopods of amoeba, but, at least for the short duration that I watched them, nothing moved. Also, I took multiple images for a focus stack (not shown) and I saw nothing "inside" t...

Help with identification
I found these in water from a pothole in the yard after it had been raining for a while. I don't remember seeing something like that before. Many thanks for suggestions of what they might be. Your images in the Magazine
May I encourage forum members to give Oliver explicit permission to use their images in the Microbe Hunter Magazine? There are superb images posted to the forum that deserve to be seen by a wider audience. I believe (and I hope Oliver will confirm this) that the members retain completely their copyr...

Amoeba (video)
This is an old video; I post it because it is colorful (in contrast to the scuticocilliate video which was devoid of color). I think this might be Thecamoeba . Objective: 20x; illumination: DIC; camera: Olympus E-P1; duration: about 3 minutes (please click on vimeo to view in HD). 108962077

Re: Scuticociliates
Many thanks to Oliver for the quick help in embedding the video.
Thank you, 75RR, for your kind comment. I'm no expert, but I think those are ciliates and they swim rather than walk or run, but perhaps someone more knowledgeable will give a more definitive answer.

Scuticociliates (probably Cyclidium sp., but could be Uronema[?] or Pleuronema[?]).

Re: 25x NeoFluar First Test
Very nice images. Did you compare it with your Zeiss planachromat? If you want to do so, I think it would be better to do so visually using brightfield with the condenser aperture adjusted for each objective, using both a low-contrast object and one with fine detail. I may be wrong, but I think it m...

Re: Objective upgrade
If in good condition, the 25x fluorite objective should be superb (watch out for delamination, though). I assume you are using Zeiss compensating eyepieces (as I understand it, this is especially important when using fluorite and apochromat objectives).
