Re: The Start-up Blues

So I prepped a slide, placed it on the stage, and turned on the supply for my nifty home brewed LED system for the first time since last spring and... POP!
The LED blew.
Sorry to hear that. Hope you are up and running soon. Any idea as to why that happened?

Testate amoeba + Synechocystis ?

16x, 95µm, Oblique, Olympus E-p2

Testate amoeba at home and active, which was nice.
Surrounded by what I think might be Synechocystis.

Those are nice images.
As usual, I've got everything against me.

Re: Pictures from an asparagus cross section II

These are very good images, but I have to say that the second one is great.
One can almost feel the shape as it curves towards the center.
Is the center area thinner i.e. single cell thickness or is it the result of image choice for stacking?

Re: A question for 75RR if I may

Hi DaveH, I bought the E-p2 second hand - body only. I was using the afocal method with legacy OM Olympus lenses at one time, mostly a 50mm with a Micro 4/3 to OM adapter. For a little while now I have dispensed with the lenses and am using the projection method using a KPL 10x eyepiece. See links f...

Here is a link to an interesting and relevant article by David Walker at Microscopy UK. It has been posted before in another thread so some of you may have already seen it. Worth another look. http://www.microscopy-uk.org.uk/mag/indexmag.html?http://www.microscopy-uk.org.uk/mag/artsep08/dw-pa-test.h...

Re: COL + Pol + offset retarder vs UGF

I just used black center stops that are slightly too small for true darkfield. Should that not be rings? I was wondering if one could make a home made Phase kit. Apparently one can. Re: COL + Pol + offset retarder vs UGF

Agree stills would be easier to compare. Still think Pleurosigma is a good choice as it is not just about resolving the punctae but about how well one can resolve them. Adding UGF + UGF with Pol is a good idea, as it would allow those who do not have Phase, and hence are not able to try COL (Circula...

Re: COL + Pol + offset retarder vs UGF

Hi zzffnn, interesting idea. Would like to test this but was wondering if using Pleurosigma angulatum, also from Kemp's 8 form slide, rather than Frustulia rhomboides might be better. The punctae of Pleurosigma angulatum can be distinguished with a 40x/0,65 objective which would be more representati...

COL = Circular Oblique Lighting. It is Oblique obtained by misusing a Phase condenser ;)

Re: I hate this 15X eyepiece

If I have a choice should I stick with the mfr's brand? That would be a yes. While I have heard it is possible to achieve decent results mixing and matching, I would leave that to the compulsive tinkerers; for a first-rate viewing experience, stick to the eyepieces that were designed for the object...

Re: Hydra (40x)

I was hoping you would post some comparison photographs. The jump from 10x to 40x is quite large, depth of field is much much smaller with a 40x objective. Lighting, focusing and contrast requirements are much higher. Perhaps you should look into getting a 20x objective - it is to my mind one of the...

Re: Hydra (40x)

You might want to do a comparison between your 40x objective and your objective that is nearest to it in magnification. Ideally you would use the same subject for both, something less amorphous would help, a diatom perhaps - do not move it, just rotate the other objective in - and use either Brightf...

Hi CCMOLA, I used a Zeiss Neofluar 6.3x and 25x. Microscope is a Zeiss Standard RA.

Yellow globules in Ostracod Part II

Decided to see if the yellow globules that have been provisionally identified as triacylglycerols were only visible if the weight of the coverslip caused a rupture of the Ostracod's shell. Leading to ...often small oil droplets coalesce to larger drops... following a squish . See charlie g's comment...
