Botanic combination stain experiment

#1 Post by MicroBob » Wed Dec 30, 2020 4:46 pm

Hi together,
here is the result of a staining experiment, showing a young lavender stem. Stained with a mix of safranine, basic fuchsine and chryosidine, differentiated with 70% etanol and a little HCl, then counter stained with iron gall ink made from gallic acid, not iron galls. I like the result as a differentiating stain that is not as wildly colourful as Etzold FCA or Wacker W2A.
Section is 40µ, made with cylinder/hand microtome and blade holder like shown here: viewtopic.php?f=16&t=7065&p=93716&hilit ... der#p93716

HI Bob
I agree. Nice differentiation and I do like the colours.

Here is another combination, first Safranine, then differentiated, then iron gall ink. This stain could use a third stain in the middle perhaps, like Chryosidie.
When looking at these stained sections it is interesting that the image changes a lot when viewed with objectives of different strengh. With weaker objectives I get duller colours. So using a lowest power objective to get all in doesn't really work as expected.

Hi together,
here I stained with safranine, chryosidine and iron gall ink. The chryosidine could be pronounced more.
