What are the specific preparations for Spirogyra to be optimally mounted in Nail Polish Mountant

Hello,
Put the sample on a slide and allow the water to evaporate. Then put some NPM on top and a cover glass. Then evaluate the result. The drying process can cause shrinkage and the solvent in the NPM might dissolve the cholorophyll of the chloroplasts. But you simply have to check the result, maybe it will be fine. In any case, do not mix a wet sample with NPM, make sure that the Spirogyra are dry.

I have found that thinning down NPM with ten to fifteen percent
Acetate, improves both placement and clarity of specimen.

Make sure that edges of slide cover glass are well sealed, again full strength NPM.
