Hi members,
A member of a small group of microscopists has aske me to show them how to make a stained botanical slide. While I feel very honoured I know need to put my thinking cap on regarding wax, stains (strength, for how long to stain etc). I thought I would give a peep at what I have done a couple of years ago.

What stain did you use on these?

If my pitiful brain serves me well the brown would have been Bismark Brown and the red would have been Carmine that is in the Field Poppy and the Cosmos. What I like about B. Brown is that one can not overstain with it.
I now need to look up some of the protocols I used a while back as I will be honest with you all I have forgotten timings for the waxing and staining.

I have a couple more that I have bought and unfortuneatly I can not read the writing on the Victorian slides.

Hi,exmarine, thanks for the botanical preps you offer here. I'm curious as to how long ago you made these botanical slides of yours? I'm curious if you can guess an age for the slides you purchased?

It's great your doing botanical microtechnique again...congrats on being an active member of a microscopy group in your area...and your doing botanical microtechnique for the group/ with the group.bravo.

JohnB. has actual schedules for dehydration times, wax infiltration times..right on to Shandon microtome processing...staining schedules...and outcomes. Then JohnB./Mr.Sonchas gives his acellerated (less time, less steps) schedules of his botanical microtechnique. Often JohnB. mentions texts from which he applies techniques...and then he shares how/where he trims ( a botanical pun?!) the schedules.

I have terrific pleasure in following these botanical microtechnique paths to microscopy....now that I aquired a group of John B.s prepared slides...I always have the garden microscopy for when the world wearies and society ceases to satisfy ( oops, sorry Louisa May Acot...err..happy birthday to you).

please vist the threads in this forum for John B.'s schedules ('shedules' it sounds like in Canada?). charlie guevara
