Is ten microns really the thinnest sections you can get with a hand microtome?

Hmm, I think you would be extremely lucky to get a good, cleanly-cut and even (thickness) 10µ section with a hand microtome - more like 20µ in my experience. A lot depends upon the specimen, how it's presented, how it's supported, the type of knife used, technique (we have a resident expert hand-sectioner-extraordinaire Called Jim in this fine forum who's images will encourage you greatly)...MicroMan2 wrote:Is ten microns really the thinnest sections you can get with a hand microtome?
A hand microtome is a great thing to have - used well it can give some really good results and fast - I'd strongly recommend that you buy one with a built-in clamp within the specimen cylinder - great help for holding specimens fast.

I would agree with John that 10 microns is a tough goal for a hand Microtome.
As John says, support of, and structural integrity of the sample material are key limiting factors. Now consider knife sharpness and technique and the microns start adding up quickly.
That said, it can still produce some fascinating results, and is well worth the effort.

John B. is too kind about my mangled sections. I not only use a hand microtome but it is home made.
Have no idea how thick they are. Some seem quite thin and I toss the really fat ones but nice thing about a TS is they offer a vertical view of cells.
