Unknown organ identification, H&E stain
I've been having a go at making my own histology slide, but I encountered an interesting problem.
First, the organ tissue was taken from a frozen whole chicken bought from the supermarket, it still had both lungs,both kidneys and what I tought to be a blood vessel from one kidney, I will post pictures of than soon. In the first image the red zone is the place from where I took this unknown organ ( I will also post pictures and how it looks). I sectioned the tissue as a frozen sample and followed a normal H&E staining protocol, lastly I mounted it in glycerol-jelly.
I took a total of 70 pictures ( the dark field image was made using 54 images at 100x magnification, and 16 photos for the bright field at 40x)
The problem is that although it shows particularities it does not seem to resemble any thing on the internet or in my histology book (their about human histology so I can see some reason behind it). So if anyone that has more experience can tell me what it is I will be most grateful, also if it is needed for a magnification in some zones I can make that too.

thanks for this posting, Raul! Your first image of the chicken organ systems is too little to assist in your excellent H&E slide preps. Chickens have no muscular true diaphragm ...so chickens have one huge body cavity from thoracic inlet...to the chicks 'vent'.
You can be shareing with us anyone of 'the bunch of grapes' chicken ovary follicles at varied states of maturation...or lung tissue 'sacs'...too little context is provided in your H&E slides...IMHO.
Since your microscopy post regards a specific and common fowl...think about a simple 'point&shoot' image of the subject with open chest cavity...then you, yourself, Raul...gain confidence in the birds 'gross anatomy'...long before you generate H&E slides of ....'viscera unknown'.
Also I suggest you include higher magnification image captures...in your current posting,,,my sense is too low a magnification to appreciate the status of all those H&E tissue cells. thanks for your thread....chickens are kin to dinosaurs more so than kin to we mammals....err...I eat too much chicken! charlie guevara

A zone ; Magnifications in order of images:100x,200x,400x,400x with 2.5xzoom,800x, 800x with 2.5xzoom

B zone + cells that were removed from the tissue (seem like epithelial cells),800x magnification of epithelial cells, 400x magnification of B zone

I believe that as charlie g named it so poetic, Viscera unknown is a lymph node. I searched thorough a lot of histology book and all indicate this. It was hard to distinguish because my section was not sectioned at the proper size of a 6-10 μ, instead with my hand made microtome I think it is around 150μm. Again with the fact that I don't know anything about chicken anatomy I can't really say for sure.

Wow, Raul...your 800X images indeed show good intact cells...but the tissue level/ the histological level is not apparent..to my old tired eyes..sigh.
Domestic fowl are so common...you can easily online get images of: chicken gross anatomy...from these images...drop the low cost duckets to buy an intact meat-case fresh chicken...you yourself should with unaided eyes see what tissues are left intact in that birds cavity. Again I remind you...these birds have no true abdominal compartments...one huge 'viseral cavity'...the processor of the bird your purchased for your microscopy...'scooped out the lungs, the GI tract, heart, liver...why do you sense at all the birds 'ovaries' were left intact for your excellent H&E slides?
Your cells show great staining...but the confidence of just what tissues you processed is not clear....we had beef +rice+cheddar cheese +green scallion with a tomato paste, stuffed green peppers tonight...I eat too much chicken! all the best...and thanks for your post and microscopy images, Raul. charlie guevara, finger lakes/US

Hi again, Raul...ooops, my bad...you never said you suspected 'chicken ovaries' as your source of tissue which you H&E stained to good cell level detail. I was mislead by your first image which has red boundary circle of a chikens ovaries,
I also am stumped by your UK purchase of a frozen chicken with the lungs left in that bird...humm. I sense this side of the pond that all the kidneys and lungs are scooped out of an intact processed chicken...some times the birds heart+it's liver+ it's neck are enclosed in a paper sack...stuffed in the scooped out abdominal cavity...this paper sack of viscera is termed: 'the gibblets'. This side of the pond,,,we use the turkeys 'gibblets' this next Thursday in a national holiday traditional meal....I eat too much chicken! all the best, charlie guevara, finger lakes/US

Hi Raul...I stand corrected...the paper sack with 'gibblets ' contains: long neck+liver+gizzard+heart .
I sense our 17.64 lb free range/organic turkey must be a tom...I'm pretty sure the two bilateral pale bean shaped organs adjacent to the bone-spine are testicles...the lobed bilateral deep red organs must the kidneys. My images are to thank you for shareing a thoughtful post on fowl tissues you carefully worked up and brought to forum...I'd like to offer your some chestnut+celery+onion+herb spiced bread cube stuffing some day! The two bilateral major nerve chords in the birds overall image also prompt my suggestion that your first see the gross organs or tissues...before H&E slide preps. Hi charlie g thank you very much, indeed I learned a valuable lesson (not to say: o, I will identify this organ after the staining ). Thank you for sharing that image, it was kinda the same that I saw and decided that the chicken will serve a even higher purpose for science (also cooking). Nonetheless I learned my lesson, but I also liked the research part of it. In the end it was a great adventure with a microscope!
