Messing with Acridine

Just got my first batch of Acridine orange. Im still trying to figure out how to stain live ciliates. I think Im adding too much and so I get a lot of orange glow on the whole slide. If anyone has experience using this dye on protist any help will be welcomed.

Acridine orange is quite toxic. Overdosing results in dead protists. I they survive the substance they may be grilled by the emitted light.

For live staining of protists I recommend 100 microliter acridine orange 0.01 % in a 100 ml culture. After 24 hours all live forms in the culture will deliver significant fluorescence while the background remains black. As you already learned, acridine orange allows to differ RNS from DNS and alive from dead.

It has to be diluted quite seriously, but in the quantity I had to buy it will be available to my great great grandchildren. :D
