Copepod and Rotifer 3D
Hi everyone,
this morning playing with a drop of water from a pond and with my new DIY 3D system, I photographed these two organisms: the first looks like a Copepod nauplius (made with a moderate 3D effect), the second looks like a Rotifer and it is made with the maximum 3D effect.

Looks like stacked images in photoshop. I will have to try the crystal method.

The first image is stacked (from a 4K Photo footage), while the second is a single shot.

Infact...this 3D setup seems to extend a lot the depth of field...also during the observation. I don't know why...

In conventional macro photography, when you increase the depth of field (for example by stopping the diaphragm very much) the resolution decreases due to diffraction. In this case, you have both depth of field and resolution! It probably depends on the angle of incidence of the light, but it's not clear to me.

Somewhere I should have a "normal" Leitz 600 series condenser ... I'll do some tests in the next few days ...
But I'm afraid the Berek's field lens should have some effect ...

This 3D aspect is due firstly to the large depth of field of 20-25x objectives, approximately 7-10x greater than that of 40x objectives. In addition, they accept without deterioration of the image a significant closure of the diaphragm under the condenser which makes it possible to further increase the resolution. The image is enhanced by oblique and incoherent lighting. I said incoherent, because, if I understood correctly, your microscope has a matte filter in front of the bulb. I obtained comparable results with a black path glued on a piece of semi-transparent plastic, placed under the condenser.
