Beautiful observations of a living diatom, I love those green hues, thank you. Can you tell us how difficult...or not difficult it is to use a live organism wet-mount slide with transmitted DIC illumination?

Is the 'tight working distance of: 60X and 100X DIC objectives" a tricky and tedious challenge to observing wetmount slideswith these magnifications?

I also ask why yout list: "60x and 100X DIC" for this video...is this diatom really that small in size that both 60X and 100X objectives capture almost this sweet organisms entire body length?

It’s not that hard.
But we need more effort to make clean and thin wet-mount slides.
Because dic is highly influenced by the thickness of the wet-mount slide at high magnification.
Dic isn't the way it works in every case. Sometimes brightfield, phase contrast is more suitable than dic.
Of course, it's just a personal opinion.
Obviously, in most cases, dic is much suitable better than phase contrast, brightfield.
Phase contrast is extremely limited. but, phase contrast shows better contrast than dic.
Of course, we need make extremely thin wet-mount slides for phase contrast.

This diatom is small.
I used 60x 100x objective with 2.5x photo eyepiece.
This vid show One-third of the area I can see through super wide-field eyepiece(26.5 mm).
