Great ones, so sharp chromosomes! I'm a little jealous :D My chromosome preps looked really crispy and weird. I guess it is DIC/Nomarsky illumination, but what is the brown stain?

Thank you all for your kind words.

I cut approximately 2-3mm off the tip of root.
Then I placed the root tip on a slide with 1-2 drops of 10% Hcl (muriatic acid) for 5min.
After 5min I added 1-2 drops of Hematoxylin for 5min.
Then I removed Hcl and Hematoxylin with paper towel and added just one drop of Hematoxylin for another 5min.
After that I removed Hematoxylin with paper towel, I added drop of water and cover slip,then I squash it with finger and paper towel.
I used Nikon CFN 100/1.25,and DIC technique.
-Reichert Polyvar
-Olympus IX70
-Zeiss Photomicroscope
-Canon 600D

Thank you, Seb28, for the additional information. I have the same objective, but cannot get anything near that quality. As I always say, the microscopist is much more important than the microscope. Thanks again.
