Chillax brand synthetique down (Polyfill)

Synthetique down or PolyFill is most frequently used as the insulation in Winter jackets. Up close it looks like this. Photo 1 Image
Soft and fluffy with lots of air pockets.

When I placed the slide on the microscope stage I noticed I had not trimmed the excess Polyfill. Photo 2 Image So I decided to try for a stacked 3D image using CombineZP.
Photo 3 is an 11 image stack with the CombineZP Pyramid Do Stack macro.
Each change in focus was a full turn of the fine focusing wheel giving an estimated depth of the edge sample of 1.1mm.

The PolyFill sample did not show much in the way of birefringence on the polarizing microscope so I moved the slide to the CxL
A close look with a 10x obj. does in fact show a lot of space between the fibres.

To get better contrast I inserted a 16mm DarkField stop. The condenser aperture was at the 100% open position.

Closing the condenser aperture to 30% improves the contrast and depth of field.

Photos 5 and 6 are good reasons to adjust the condenser aperture and try DarkField illumination.
