The microscopic study of bone (self made slide)

#1 Post by Raul » Tue Mar 15, 2016 8:49 pm

Hello all, I wanted to share some images from a new project of mine.

The good donor for this sample is an unknown pig.
I will also post pictures of the procedure for the processing of the bone if anyone is interested.

Hope you enjoy it.

Thank you very much billbillt.
As promised the protocol is the following:
The samples (left is a chicken femur and right bone unknown origin from a pig)
Image
First I tried the femur but was only able to section it in half (still trying on it)
Image
For the pig bone I used a scalpel and tweezers to remove a small piece
Image
After I used fine sandpaper to grind and at the end polish the sample
Image
The sample was 0.132 mm thick when I decided to stop because it seemed to fragile to polish more
Image

I mounted the bone in glycerin because of its high RI and the fact that glycerin is used as a clearing agent for bone (making the sample more transparent)

The sample showed great details using different filters and didn't require staining

Brilliant.
Always good to see people pushing out the sectioning skills.
Lots of work, nice result.

What about a tooth?;)

Thank you billbillt and billben74 i appreciate your feedback very much. billben74 yes a tooth is a very good and interesting idea, that would be a future project as soon as I find a donor for that (hope it won't be me because I have my permanent teeth).
