16x Objective, 50µm width, Brightfield, Darkfield and Offset Darkfield Stop, 8,6 and 9 image stacks in Photoshop

These are still shots taken of the same subject as in the Spirogyra and Trachelomonas video.
Curious how the internal spiral has contracted in the Offset Darkfield Stop image. All three image sets are next to each other in the "virtual film roll" (i.e. consecutive image numbers), though the Offset was taken last.
Order was Brightfield stills, Darkfield stills, Darkfield video and then Offset.

Not exactly sure what you are referring to. Photoshop does sometime outline "areas" when it blends images.
If you open the second image in Photoshop and use the Magic Wand on the background they will show up.
These areas are not normally visible at usual monitor settings.
