VERY glad to be back!

#1 Post by McConkey » Wed Jun 22, 2016 3:22 am

Good evening all!
I returned home last week from a few months away due to work and could not wait to start up the microscope again! After unpacking everything i re-packed and took off into the mountains! A nice weekend away with the wife was the official reason, however on our hikes i managed to grab a few samples from here and there and bring them home!

The Rocky Mountains are only a few hours away and many of the lakes within the mountain ranges are fed by melting glaciers so it seemed like a perfect opportunity to get some less than typical water samples! I grabbed 4 water samples from different lakes at an average elevation of 1200m above sea level and 1 moss sample from a peak of the Mt. Edith Cavell Meadows at roughly 1700m above sea level. With the air being thinner at these elevations and the water being glacial fed, i wasn't too sure if samples would be different from the average pond water!

Here are some of images from the first water sample!

Unfortunately i was enjoying my time back at the microscope a little too much and wasn't paying attention to the objective used for these images. All are stacks.

Thanks.!

Welcome back. Sounds like a super camping trip.
Very nice series of images. First one is quite striking.
Unfortunately i was enjoying my time back at the microscope a little too much and wasn't paying attention to the objective used for these images.
I know what you mean! I keep having to remind myself that every photograph has two steps - make a note and press the shutter button!

Good to have you back. Wonderful images of a nice variety of critters. Any idea what the first one might be?

Thanks guys! It's great to be back! I have been trying to catch up on all the posts you guys have been making! Some really excellent images over the last few months! I'm especially loving the new interest in Diatoms and all the images been produced!

Gekko - I don't know if you noticed the file name of the first image "Blob stack" ahah this is the extent of my knowledge on the specimen! I'm afraid I don't really know much about any of the images I took.

I'm going to peek at another sample tonight and see what's lurking!

:) I hadn't noticed the image names. Your images are all excellent.
I'll take a few guesses (hopefully someone who knows will confirm or correct my guesses):
#1 A beautiful blob (cannot do any better :) )
#2 Spirogyra, a filamentous alga.
#3 Filamentous alga, but I don't know what genus.
#4 A dinoflagellate, probably Ceratium.
#5 A ciliate (I don't know what it is).
#6 Maybe a desmid, perhaps Closterium?
#7 A diatom (I cannot identify it further).
#8 A green alga, perhaps Scenedesmus.

You are definitely making that 20 behave, Karl. Are you sure #4 isn't just a medieval celtic clan symbol?

Thanks for identifications gekko! much appreciated!

Aha #4 is my family crest Phil, tried to slip it in without anyone noticing!

Thanks Seb!

I've noticed that a lot of the time the specimens have a colourful glow around them. Sometimes it distorts detail, I've tried adjusting the height of the condenser, angle of the light via the mirror, aperture that the light comes through...a whole bunch of things but the colourful glow remains. It's very apparent in the image of the diatom. Is this a common issue with a simple solution?

Thanks again for all the kind words! Hoping to grab some more images this weekend!

Was the diatom picture taken in phase contrast? It is normal for phase contrast images to have halos, I think.

Good variety of subjects and very nice images.

#4 is a Keratella rotifer (genus). Possibly Keratella cochlearis... but I'm always hesitant to go as far as exact species.

#6 is definitely a Closterium. There are many species with this long skinny "bow" shape. (Closterium acutum and Closterium keutzingii look close... but again there are many species that look like this, so these are merely suggestions)

Yes, #4 is Keratella Rotifer. I wanted to name #4 as a Rotifer, but I could not even remember the genus. Charles nailed it!

Gekko - The diatom was phase contrast! That's good to know it's a regular thing! I won't worry as much now!

Thanks for the more specific identification Charles! It helps a lot!

Fan - really appreciate the links! I love seeing the same subject from different angles. It's incredible to see how different these guys look when they're moving around!

Many thanks for the correction. I would never have guessed. n
My apologies to Karl for my unintentional misleading identification  .

Haematococcus? What's that? :) . You may well be right! The intense yellow color fooled me, I think.

I think that's an artifact of the illumination method.
