Larger Diatoms Just For Fun
Just having a little fun with the microscope and camera, mostly fooling around testing a trinocular head I refurbished - finally got a black one to match the rest of my Zeiss WL stand and had to clean and re-lube the innards to make it work like I want. This is a Raymond Hummerlink slide, don't know much about the sample other than that, except to say it's rich in Pinnularia and Nitzschia.
First image is a Pinnularia through the dreaded 40x/.65 lens, DIC - whaddya think, 75? Second is Pinnulria through the 100x/1.25 lens, DIC, 1st order red compensator; not a great image, but I liked the colors! Next two are same through the 100x/1.25, DIC. Finally, a Nitschia that was flat enough to actually do some good with a single frame.
I really, really need to conquer stacking...

I am certainly no DIC expert, but they all look great to me. Outstanding resolution on all.
I would certainly be pleased if they were mine.
The new head is like a crown on royalty. That is one sweet rig.

Wow. Even better looking diatoms now Kurt!
This kind of quality has to come from mastering of both the microscope components (objectives + condenser fiddling ) and the way you set up the photographic system.
Sorry if you had explained this somewhere on this site already, but are you using afocal with your setup?

These are really nice. I'll reserve judgement on the 40X (always best to make a judgement of subject that "fills-the-frame" more). The 100X looks really good.
On the second image... which is excellent... you have too much of it's splendor buried in the dark tones. Check out the histogram. A simple levels adjustment brings it to life.

Beats me what it's called. A dSLR (Canon T3i) is mounted to the trinoc photo tube so I'd say no, but there's a 2.5x photographic eyepiece inside the tube so maybe it is? Never mistake me for somebody who knows what he's doing.hb2sbdb wrote:...are you using afocal with your setup?
Not the first time I've been fussed for too-dark photos. I like 'em dark. I know I need to knock it off, but I just can't stop. Must have goth-emo tendencies. No stacks, that's the problem! All the above are single frames. My efforts at stacking with Combine ZP and Picolay have resulted in pure #@&^%. I am considering trying Zerene (aye, blame it on the software!).McConkey wrote:What are you using for stacking/how many images were stacked?

For single frames these look really good! With the level of detail i thought they were stacks! Great job!
I've had mixed results with picolay, some great some not so! I havn't heard of Zerene but i'll check it out!

In the PM to c-krebs I invited him to perform a demonstration of post processing on image 2, as many of us are sure to be interested in his opinions on the matter. But as I have had other commentary on it, I hereby extend the invitation to anyone who wishes to play with it. Shoot me an email and I'll send full rez original file of the image.

I took up Kurts invitation and requested a full size image to play with. With his permission i'm posting them.
Please keep in mind this is an alteration based on my personal taste and is in no way a critique on Kurts awesome work. Just a show and tell of how images can be altered in post processing. Diatoms are the same in both images...just one has a lighter background.

Kurt, I use Photoshop CS5 but what I did can be done with any version of photoshop.
Reduce noise:
Strength - 9
Preserve detail - 60
reduce colour noise - 90
sharpen detail - 25
Sharpen - 30 x 30
Selective colour:
Blacks - Cyan +7
Yellows - Yellow -100
Cyan - Cyan -100
Shadows/Highlights:
Shadows - 0
Highlights - 5
40% Burn Tool on Diatom
Hue/Saturation:
Hue - (-11)
Saturation -43
- To turn the background to a neutral gray i created a duplicate layer (named gray layer) and with the selective colour adjustment tool, under black...turned down the black until i had a tone i liked.
- I then applied a 15pixel Gaussian Blur to the "Gray" layer and applied a black layer mask (revealing the original image below)
- Using the white paint brush at 90% opacity i "painted" in the gray background on the black layer mask.
- When i got closer to the diatom edge i reduced the opacity to 40%...and then when i got even closer reduced it to 10%
Again, this was just a fun little play around with an already superb image that didn't need "improving" or "altering" in any way, shape, or form. Just a demonstration of post processing.

Opinions no doubt vary, but here's mine (since it's my image): Post processing is part and parcel of digital imaging, and is not "cheating" in any way, shape, or form. I'm speaking of cropping and adjusting levels and such, not cutting and pasting to make "photo-chopped" images. While I very much appreciate the kind intent when you say my image needed no "improving" or "altering", I gotta say yes, actually it does, and so does any image as far as I'm concerned. When you have a whole world of adjustments to make, there's simply no reason to accept an image straight out of the camera as the best you can possibly offer.
I'm not sure I've taken a picture yet that couldn't be improved by tweaking saturation or contrast, or maybe sharpening, or whatever helps it. And it isn't just a modern thing, top photogs used all sorts of "darkroom magic" to put a polish on their images back in film days.

If anyone else cares to take a shot at it, the offer's still open.

Thanks for the reply, 75 - not only was I hoping for more post processing input, but the sharpening demonstration helps bring out some detail that got me all hung up on this particular diatom lately. Some weeks (months?) back I started asking what makes Pinnularia and Navicula so different that they're a whole different genus, especially when some species can look so very similar to those on the opposing team? Well, the answer is (in part) in the stria. Seems Naviculae exhibit areolae in the stria, while Pinularia stria are chambered (thanks exmarine for the clue!). Okay that's a good indicator, just look for the longtitudal lines created by the presence of chambers. But what in the world is going on with chambered stria?!
Seems like every week I'm obsessed with a different diatom, but that's what makes this so much FUN!
