A couple pictures of a Closterium

#1 Post by Pat Thielen » Wed Apr 19, 2017 5:13 pm

Here are a couple photos of a Closterium; one is taken using oblique phase lighting and the other is dark field. I'm still working on the oblique lighting technique, but I've liked what I've seen so far. I did try taking a stacked image of this guy; I took ten photos total but I didn't like how it came out once I had processed it (it just didn't look real and the edges of the Closterium didn't come out right). As always, let me know what you think of the images -- Any advice for improvement is always greatly appreciated.

I like them both. Could you see the crystal movement in the vacuole?

Very nice, Pat!
Have you tried oblique alone. Or using 100x or 40x phase ring with 20x objective, for circular oblique light?

I certainly did! :) Also, this guy did a slow ninety-degree turn as I watched. It was really quite interesting.

I haven't but it does sound like an interesting idea. I'm not sure if I can do oblique alone with my microscope; I'll have to do a bit of research first. Thanks for the suggestions!

Thanks for the link -- I'll give this a try when I get a chance. There are some really interesting lighting techniques to try out!
