Grape Stem

Transverse section (TS) of a grape stem stained with red food colouring. First image is with the 4x Obj. and gives an overview of the cellular structure. Sorry about the blurriness of the image.

Second image is with the 10X obj. and begins to show detail of the Secondary Xylem and Cambrium.

Third image is with the 20X Obj. and offers more detail of the Cambrium and Xylem.

Two and three are oblique lighting hence the 3D look to the cells.

Hi Jim,

I think you did a great job with these.. Your oblique lighting gives a great 3D effect..

Couldn't agree more than I do with Bill - lovely set of good sections. The differentiation gleaned from the use of the red food-dye is very satisfying indeed - many tissue types distinguishable. True too that a view 'down into' the xylem vessels is seen!

Great colours as usual with your technique too Jim. Thanks again for a nice set and a great demo of just what may be done with a good hand-section by a skillful technician.

Have you tried any of the other food colors?.. Blue, green...etc..?

Yes but I didn't think they revealed as much detail as the red. I should try again.

Might not have been the food dye's fault as much as the person doing the staining :)

Now then Jim, have you tried standing the freshly-cut stems in the staining solution with a view to the stain being taken-up by the Xylem - maybe - I know this will work for some plants....

Then section and 'see what you have' - may be worth a try.

Nice sections..20X I sense with no tissue distortions, what sort of blade,Jim T? My dear concord grape vines are doing sweel on their wire fence this spring..what part did you section..store purchased grapes on stems..or from your home garden?

Charlie, a razor blade with tape on one edge so I don't cut my fingers off. Secondly, store bought grapes on stems so a little woody.

Cranberries in the Finger Lakes :o
