Some Algae DNA

#1 Post by kinase » Mon Jun 12, 2017 2:03 pm

I neglected to take white light photos, probably because this dye bleaches fast so I was focused on that. This was stained using SYBR green, we're at 40x. These cells occur in diploids. You can also see what I believe to be a nucleolus with some odd ringed structure around it. Not sure what that might be, I might have a go on a confocal and check it out. These have many plastids, you can see that each one has it's own small genome.

This one is the same, except you can see that the cell in the bottom left is working on finishing anaphase. It's possible that some of the cells that look less like two individuals are working on cytokinesis and nuclear envelope reformation.

Very nice! I have never seen these structures before. Do you know how I can get hold of SYBR Green? Any good sources which sell to normal microscopists and not labs only?

Ouch....

Yeah you don't buy 50mL of any kind of fluorescent stain. That amazon link has $40 shipping, and that's still oddly cheap. Thats probably for making a lot of DNA gels or some other application, maybe lots of qPCR. I'm not sure that you can just buy the stuff. It's typically 10000x concentration, i.e. it needs to be diluted 10000x in a new solution to be at it's recommended concentration. So I barely need a single microliter, usually just sucking a visible amount up is probably way too much to stain less than a mL.
