Cross section of aneedle of Picea spec.
Cross section of aneedle of Picea spec.
Dear all,
motivated by the section of a pine needle shown by Rod (rnabholz) I tried today the same.
Believe me - its a real challenge.
The section was stained with Wacker FCA.
In picture 2 I try to explain the details, unfortunately only in German (my English is too poor )
Any comments are welcome, thank you for your kind attention.

Perhaps it is my fresh experience, and less than optimal results speaking, but this is ABSOLUTELY FANTASTIC!!
I had planned to take another run at this subject tonight, but your shot may have defined the genre for all time.
Anything I might post will look like a mud pie now! ;^)
Would love to hear more about the technical details.

Hello Rod,
thank you for your nice comment.
Most important is to try again and again, also my first attempts were really disatisfying.
Most of my equipment is homemade (microtome, razorblade holder).
To achieve this image I did several dozens of sections, only a few of them were thin enough (ca. 50microns) to allow staining.
Go ahead this way, you will see, your sections will improve day by day.
Warmest regards
Manfred

Hey you've made a great start yourself - I've not even produced anything that's not too bad to post and I've beenrnabholz wrote:Perhaps it is my fresh experience, and less than optimal results speaking, but this is ABSOLUTELY FANTASTIC!!

trying for a couple of months now. Your technique will definitely get better the more you practice.
I use images from others as my targets to achieve and am slowly making progress too.
Have another go, and then another - it's a great feeling when you see your own progress, especially when you
move into making permanently mounted slides for your own collection.

Manfred, (or anybody who would like to help)
What can you tell me about the Wacker FCA stain you used? I had not heard of it.
Thanks

Re: Cross section of aneedle of Picea spec.
Rod,
it is also named "Etzold FCA" and is a mixture of Fuchsin, Chrysoidin and Alcianblau (sorry but I don`t know in English).
It is usable for all plant sections and gives nice results.

This stain certainly does a good job - I wonder what the 'UK version' is sold as?Manfred wrote:Rod,

Manfred, this is absolutely superb (which is always the case with your photos). Gorgeous tapestry! Thank you for posting it.
