My first feeble attempts to stain & mount onion epidermis...

#1 Post by mrsonchus » Thu Feb 12, 2015 12:47 am

Here are several of my first rather poor attempts to stain onion epidermis
with Safranin and Fast Green stains (seperately).

The samples were kept for several days in 75% IPA then stained for 2 hours. They
were then taken through alcohol/DIW series down to DIW only, then mounted in
glycerin and sealed with nail-polish. A quick and dirty procedure I know but
I'm learning with every horror I produce!

Sorry these are a bit messy-looking, my camera is not giving me anywhere near
the image quality that I see through my 'scope. I'm saving to buy a DSLR to mount
instead - hopefully a jump from 2mp Toupcam to an 18mp Canon will help with
photomicrograph quality?

Hope you like them

John

Thank you, Walter Dioni inspired me to try the onion epidermis, have a look at the website and his
contributions - you'll soon get bitten by the bug!
