Hi, I've just taken this 6-picture stack (stacked with Toupview) of an onion epidermis stoma through
a 60x objective...
I'm pretty pleased with the level of detail, I like the nucleus detail with it's nucleoli bottom left too. The Canon is definitely a big step up from the 2mp Toupcam.
Here's a 4-image stack through the 40x objective
Schlembergera with pollen-tube starting to emerge from pollen grain.
Sonchus pollen without any background cleanup....

The 60x onion image is good but I see more detail in the 40X. Too much enlargement???

I agree, but the 60x is very sensitive to mount quality and the proximity of specimen to underside of coverglass, also the refractive index of the mounting medium and the overall thickness and flatness or the slide..... (I've just been reading about the importance of coverglass characteristics as objective power and NA rises, by Mr Kim)...JimT wrote:The 60x onion image is good but I see more detail in the 40X. I also think my choices of focal planes for the constituent pictures of the stacks was definitely sub-optimal!
Basically I made a right old mess of it and didn't do the 60x any justice at all - through the 'scope it's a real performer.
I'll try another demo with some better slides and an improved technique (I hope..).
I'm going to post the Camera to 'scope mount details as soon as I've got some pictures of the parts together for the post.
