640X Dark Field Sample
640X Dark Field Sample
I have 16X Eyepieces that came with the microscope, it is not in use much. When I try Dark field technic with simple patch made from transparency with black dot glue in the center. I can use Dark Field patch up to 40X Objective.
With 10X Eyepieces it give me 400X magnification. But I cannot use DF patch to work with my 60X Objective. For more magnification, I try to use 16X eyepieces, it give me 640X magnification in DF mode. The result is very good as show in the photo.
40X Objective x 16X Eyepieces (640X)
Afocal technic using smart phone take photo from eyepiece.
Resize and Unsharp mask.

Re: 640X Dark Field Sample
When I swing the DF patch in the filter tray off center a little bit, it give me Oblique Illumination picture, which is also looking good.

Re: 640X Dark Field Sample
That first picture is nice but the second one really impressed me. Oblique Illumination certainly defines the detail to my eye. What type of lamp are you using in the microscope, an LED, Halogen? And what do you set the camera's light setting to, auto?
Re: 640X Dark Field Sample
Very nice images! They show that it is not necessary to own the most expensive equipment to acheive good results.
The darkfield spot works very well in your case. The 40:1 objective tends to be the limit for this technique and it doesn't work on all microscopes that well.
I like oblique lighting very much too. In the scientific world it is not so well regarded because it introduces artefacts into the image - here we amateurs have a real advantage!

Hi coominya,
Thank you very much for your comment. My microscope use LED and I use camera setting to auto in my smart phone. I focus by seeing on the smart phone screen. The smart phone is Samsung Galaxy S6.

Re: 640X Dark Field Sample
Hi Bob,
Thank you very much for your comment. My set up for DF is not work well for 40X Objective at the first time. I found that the Condenser is not properly align with the Objective.
So I have to loose the Condenser mounting screw a little bit and centering it to the Objective. Then it work well as show in the photo.
The Oblique is looking good, like you said it is real advantage to amateurs

Very nice images. Which LED is it - which color temperature?

Re: 640X Dark Field Sample
I use an samsung S5, but freehand, and am amazed how the focus always adjusts perfectly. I have a tungsten lamp in my scope though and use the incandescent setting on the camera. It's ok, but not a true rendering. Your images look very natural.Suphot wrote:Hi coominya, I focus by seeing on the smart phone screen. The smart phone is Samsung Galaxy S6.

Re: 640X Dark Field Sample
Hi coominya,
You may need some support to your smart phone to easier focus. I use smart phone zoom to enlarge the picture in screen and focus it. Then zoom back to 1X and take photo. It is a hit and miss (some photo sharp, some photo not so sharp).
My LED is white and look a liitle blue, so I think the color temperature is more than 6500 K. may be 7500K.
If you have blue filter, put the blue filter in the light path. It help smart phone to set better auto white balance. You may try.
Cheers,
Suphot.
