As always - such fine, impressive details ! is the diatom alive, or cleaned?
Re: Pleurosigma + 2 Anisonema vitrea
Thanks Hobbyst46,
not cleaned but it is from a marine sample that is a few days old (which pongs a bit) - not sure if there was anyone home at the time.

Outstanding image!
If that's P. Angulatum, that's one of diatoms I like to view on Klaus Kemp's 8 form test plate.
For me, the pores (punctae?) are less easily resolvable, but that's because the highest N.A. objective I regularly use is 1.25, and the only objective I have that's higher, at 1.30, is an antique Leitz 1/12 Oel.
What was your diatom specimen mounted in?

Re: Pleurosigma + 2 Anisonema vitrea
What I meant to say, was that 1.4 N.A. objective must be oil immersion to have an N.A. that high?
I never thought about using oil immersion on a temporary wet mount, but you've shown it's definitely possible.
Did you oil the condenser to the slide also?

Sorry, my mistake. Yes oil, top and bottom! In for a penny as they say ...desertrat wrote:What I meant to say, was that 1.4 N.A. objective must be oil immersion to have an N.A. that high?

Only downside to using oil on a wet slide is if the subject is near the edge ... it can seep in and lift the slide.
Not something you want with the Planapo 63x/1.4's working distance
