Nice clear video. You have lots of interesting things going on besides the amoeba.

Some tips if that is ok ...

5:25 min at 1080p is a lot of bandwidth. You should perhaps edit for highlights

What objective did you use? 10x?

Might be an idea to get a little closer. If you find the 40x too much (it works best with fairly static subjects) you should look into getting a 20x or 25x.

A practical nosepiece arrangement is 4x - 10x - 20x - 40x Leave the 100x oil out for now. Keep it in an objective "pot" until needed.

I agree about the length, all the videos I've created so far are all a bit too long. The file is almost 1 Gig at 1080p. Is there any way of forcing embedded videos to load at low resolution?

I use 4x and 10x with the camera at prime focus. This was shot at 10x. The 40x has next to none working distance (it is spring loaded) and when focused is usually in contact with the cover slip, which messes with wet specimens. On less expensive scope like mine at 40x you start to lose contrast and the image starts to get soft. It needs to be replaced with a longer working distance model. A 20x would probably hit the sweet spot.

Stage one is to edit for highlights. Stage two is to reduce file size with Handbrake.

The 40x is touching the cover slip because you have either too much "stuff" under it or too much water.

You can reduce the amount of water by putting some absorbent paper to the cover slip edges.

The less things you have under the cover slip the easier it is to see them. Try with an almost clear drop. In this case less is more.

Impressed. I ran it through Handbrake, same codec, same resolution and reduced the file size by 2/3.
