Blood
I made this video of a drop of blood donated by my girlfriend. I thought I would see more or less round red blood cells but what I saw is very different from that. What is visible in this video? Is my girlfriend an alien?
Re: Blood
Is that a petri dish? It rather looks as if you focused at the wrong depth.
It is best practice to start with a lower magnification objective (find subject - apply Köhler illumination) and then work your way up to the one you want when the objectives are parfocal, as yours are.

She is with you ;)Is my girlfriend an alien?

Re: Blood
No, it is a drop of blood on a glass strip with a cover slip, magnified 500x. I also tried lower maginfications and tried other focus posititions, but without any change (except for less sharpness). I will try again today. Is that a petri dish? It rather looks as if you focused at the wrong depth.
Re: Blood
I can actually see the platelets, they look to be at a lower focus level. No, it is a drop of blood on a glass strip with a cover slip, magnified 500x. I also tried lower maginfications and tried other focus posititions, but without any change (except for less sharpness). I will try again today.
Start with your lowest magnification objective - move the stage up and down until you see something pink!
You also need to clean your cover slips :)

Re: Blood
Wet blood samples can be less than exciting. Look up how to do a smear and stain, then you can see some interesting things.

Re: Blood
Maybe there was not enough blood on the glass. I made a new sample, with a drop of my own blood. I think it looks better.
Re: Blood
Well done.
Hand holding a camera is quite difficult. Have you decided how/where you will attach the camera and whether you will use direct projection or afocal?

Re: Blood
I have ordered a cheap adapter for direct projection (without camera lens). It will take some time to arrive though: somewhere between January 8th and Februari 8th The video above was made afocal. I have much experience in afocal bird photography through my Swarovski spotting scope (named digiscoping), so I am thinking about making an adapter for my microscope for afocal photography as well. The eyepieces of my Orthoplan though, do not fit into the vertical camera opening, so I will have to place the camera behind one of the visual eyepieces. Or find an eyepiece that does fit.
Re: Blood
I believe the Orthplan takes 30mm eyepieces but the phototube takes the more common 23mm.The eyepieces of my Orthoplan though, do not fit into the vertical camera opening, so I will have to place the camera behind one of the visual eyepieces. Or find an eyepiece that does fit.

Re: Blood
That's a very nice video! Is this also blood between a glass strip and a cover slip?
To get a video on the forum, click the youtube button above the message window, which will give [yoube][/youtube]. Then paste only the last part of the video address between the two 'youtubes' , so for your video only the text: zUnwnt6VW5U. That will do it.

Re: Blood
I'm sure if your follow this procedure you'll have better results.redflanker wrote:thank you, Is that you in video?75RR wrote:How to do video: https://www.youtube.com/watch?v=ROOPOqsiqxE
your first video looks to be focused on your cover slip. the second one looks OK, but the blood is to thick. If you try a drop of blood with a drop of water and mix with a needle, might work to thin things out . I don't now I've always done smears then stain.
