Planapo 63x/1.4, DIC, 100µm length, stacked in Photoshop, Marine sample, Alboran Sea.

Girdle view. Upper and Lower valve overlap can be distinguished.

Fantastic image ! they are so nicely distinguishable !
Do you think that this type of overlap can be identified with phase contrast ? (of any diatom, not necessarily this one)

David Walker's article Diatom Pleurosigma angulatum - a versatile 'demonstration' subject. demonstrates the NA (Numerical Aperture) penalty that Phase levies.

I think you would therefore need a Phase objective with a very high NA to manage it.

Mind you, he also shows that using a Phase objective in Brightfield (and I imagine even more so with oblique) retains or at least pays a smaller NA penalty.

I am fortunate that the diatoms in the Alborán Sea are photogenic

Map for those who might not have heard of the Alborán Sea or can't place it.

Hornblower fans might recognize some of the landmarks.
