Very nice Roldorf, quite a bit of detail to see there! Plants never cease to amaze and delight me - always a new detail, not always microscopic - always a new angle. Nicely imaged, thanks for having a go. Now my friend - we need more while the anther's hot!

Currently working on a Daisy will post when done. I missed a lot of the early flowers last year, most had already flowered by the time I started with pollen.

Hi John, just dry under a cover slip. Stacked with Combine ZP. Thought I would give the program a try. I like that you can run all the stack options in one go and then choose the best result.
Apple Pollen
Hi all, took a couple of flowers from our apple tree this evening and made another quick GJ slide - no high-res oil 100x tonight - no time.
A few images of Apple pollen grains in glycerin jelly mount with fuchsin stain and coverslip,
Focus at equator to sharpen edges,
Another aspect,
Always useful if a swollen/hydrated grain (the larger) is able to be seen alongside an essentially dry grain - the smaller one here seems not to have swollen but still taken-up the fuchsin...
An attempt at a crude stack and sharpen, but tomorrow I hope to have time to take some high-res 100x oil with IF550 filter to hopefully reveal some detail of what appears to be a reticulate exine. After a few days I also notice that the resolution from these glycerin jelly slides improves, likely as the jelly dries a little....
That's all I have time for tonight, I've had a busy day! Back tomorrow with some higher res images with luck! I may also add a few phase 100x oil images tomorrow - perhaps we'll catch a glimpse of a nucleus or two/three!

Forgot to mention that these images were mounted using glycerine jelly with metheline blue (I'll have to get a few more stains) and were a day old.

Excellent images! I really like the 1st - the micro-pores (for want of a better term...) are visible. Great revelation of detail.
I must say it's great to see folk exploring pollen! You've made a lovely job of that mount old chap, more please!

When I look at the images of pollen I often wonder how it is possible to be not allergic against them. It can't be agreeable to get these into the eye!

This has made me look at dandelion pollen more closely and see a lot more structure. Up till now I thought it was just a furry yellow ball.

Fantastic job. Congratulations !!!!, I have never worked with a 100x objective and I do not know if a stack of 2-3 images could have been made for a full focus, because the last image where you have inverted the images with a stack of about 3 images would have been wonderful with good processing, it could even have been colored SEM (it is very easy to give SEM color too). Can you try stacking?

I have been reading the forum and learning for a little time now and Mrsonshus has shown some inspirational pollen (and other) posts
and it has inspired me to dig out an old microscope ( that I was given a long time ago, along with a 'box of bits', that I did not achieve very much with, old eyesight / poor technique/ poor workman blaming tools !? whatever, it's a long story ! )
So without further ado :-
My stack of 8
(I had to learn Picolay as well as my first foray into photomicrography)
of dandelion pollen.

These look like a pretty good start to me, nicely done.
The overall 3D nature of the grains is definitely visible - keep it up, you'll be surprised how quickly these techniques can be picked-up.
I've been taking a very quick peek at some perennial-cornflower and garlic-cress grains this afternoon, but the results (in immersion oil) were so bad I've nothing to keep let alone post! Next-up for those two is good-old glycerin-jelly I think!
