Fragilaria
Fragilaria
I found these diatoms in the brown scum thats covering submerged stones in a river. The scum is absolutely loaded with diatoms, strongly recommend checking it out for all the diatom enthusiasts out there.
Best regards

Great photos.
Where is the stream and what do you think was the water temp?

Re: Fragilaria
Thank you all for the kind words.
That would be the Danube river in Central Europe, the temperature was probably around 15-20˚ C. I think in general any river will do, just look for the brown slime thats covering submerged objects. Diatom diversity serves as a bioindicator for how polluted the source is, the more pollution the less diversity but not necessarily less diatom density.DrPhoxinus wrote: ↑Wed May 13, 2020 5:26 pmWhere is the stream and what do you think was the water temp?

Re: Fragilaria
Thanks for the information Wes,
Years ago I worked on the karyotype of the European Mud Minnow, Umbra krameri, endemic to the Danube and Dniester river drainages. I think it is threatened now. It has 44 chromosomes and the North American Umbra limi has 22 metacentrics, probably Robertsonian fusions.
Banarescu P, Taisescu E, Joswiak GR (1983) The karyotype of Umbra krameri (Pisces: Umbridae) in relation to other members of the suborder Esocoidei.
Now microscopes are a hobby.

Re: Fragilaria
Thanks Hobbyst! The first phase contrast image was post-processed, I fixed the levels and gave it a tiny amount of unsharpening, tweaked the color curves a bit but the most significant modification was the clear background (its a separate solid color layer that reflects the averaged color of the natural background). The second phase contrast image was virtually untouched, only the levels were pushed by a tiny bit but nothing more (post-processing didn't make it look better).
Fascinating, does the European Mud Minnow have only telocentric chromosomes that fused into the metacentrics of Umbra limi? What is the evolutionary relationship of the two species?DrPhoxinus wrote: ↑Wed May 13, 2020 6:34 pmThanks for the information Wes,

Re: Fragilaria
I agree but the Zeiss Neofluar 40/0,75 Ph2 is also not too bad
Great Photos Wes!
Re: Fragilaria
Thanks.
Indeed, this morning I scraped (with a toothbrush) a stone from a tiny temporary stream and found diatom treasures - hopefully will not disintegrate upon cleaning !
The 40X/0.75 Neofluar Ph2 is my bread and butter tool.

Thanks Jochen. Its the only phase contrast objective I have and it gives nice results. Would be interesting to compare to higher end 40x phase contrasts objectives one day.ImperatorRex wrote: ↑Wed May 13, 2020 6:50 pmI agree but the Zeiss Neofluar 40/0,75 Ph2 is also not too bad
Great Photos Wes!
I was thinking of planting a flat piece of smooth glass and going back to it some weeks later to scrape off the slime layer with a credit card. If it doesn't capture too many sand particles it could be a great source for permanent slides.
