This cotton stem was taken from a prepared slide.
Looks very nice though.

Beautiful - STEM OF COTTON PLANT 40X
Re: Beautiful - STEM OF COTTON PLANT 40X
Thank you for showing this image!
Did you slice, stain and cover it yourself?
The image is perfectl sharp at the border, but somewhat unsharp in the middle. This is probably caused by your objectives not beeing planar types". You might improve this by stacking severals shots. I think you are using a smartphone? For some smartphones "open camera" app offers fixed focus photography. You would just change the fine focus a couple of µ for every shot.
The sharpness at the border is about perfect, a photoshop expert would probably clean the background and adjust the white point. So congratulation, nothing serious to complain!

Those dark holes are interesting. I assume they are used to transport nutrients in solution up the stem. They are surrounded by clusters of elongated cells that I suppose are there to strengthen the passageway and stop it collapsing?

If you are good at guessing you don't have to know much!
I think you mentioned it somewhere that you use a phone.
Stacking requires a fixed camera focus and a fixed camera. If you can acheive this with your phone you could of cause use stacking.
