Canon M50 to Zeiss Axiovert 25

I need some help to adapt a Canon EOS M50 (APS-C sensor with 22.3 x 14.9 mm) to an Zeiss Axiovert 25 CFL Microscope. A Zeiss V25 1.0x C-Mount video adapter (1108-963) came already with the microscope. Adapting the camera with a simple EF-M to C-mount adapter works not great because the poor sensor usage. This is obvious since this adapter is made for special microscope cameras.

Question:
In order to fit the Canon M50 is it possible to reuse the existing 1.0x camera adapter and add another lens to get an enlarged picture section or would it make more sense to get a new and different camera adapter. The final goal is to take fluorescence pictures so optimal light yield is desired.

If I need new adapter does anyone have a recommendation since I can't find the Axiovert 25/40 camera port V25 (35 mm diameter) adapters on the aftermarket. The most adapters that I found where mainly 23 mm or 30 mm for eyepiece replacement or other port diameters. Can these common eyepiece adapters used on the camera port, e.g. with adapter ring from 30mm ->35 mm?

Since the microscope is actually almost 20 years old the original SLR adapter (see picture below) fits to analog cameras/cameras with a full format 35 mm sensors. The compact digital camera adapter may be for another sensor format also, but I am not sure.

Unfortunately I am not very experienced with the details of optical and imaging systems on microscopes. Some guidance would be much appreciated.

Hi,

for your type of sensor this might be the best option you can get.
The T2 / 2.5 will crop to much of your frame unless you change your camera setup to full frame.

Don't think there is any or much difference between your video option adapter and the one for digital compact cameras regarding size on sensor.

Fitting another lens in front could help, but will probably not help much on improving picture quality.
If you don't care cropping all of your images yourself later in post processing (you could easily batch process them), i'm pretty sure the image quality you get from your setup is far sufficient.

The EOS M50 pixel pitch is 3.7 um and it looks like Zeiss sells a variety of "Axiocam" models typically from 5 to 12 megapixels with pixel pitch in the 3-5 um range. This may indicate there is not much room for improvement since the EOS M50 is already sampling the image with resolution comparable to the dedicated ~1" format Zeiss cameras. In other words, ignoring the extra pixels and cropping as Stefan suggests, you end up with something pretty similar to the dedicated Zeiss cameras. Cropping to 1" format (13 mm across) for example would leave 24 MP * (13 mm / 22 mm)^2 = 8 MP.

Thanks for you response, doing some batch post processing should not be a big deal. Since an overlay of brightfield and flourence pictures will be done anyway.
I will have to work on the overall workflow anyway.

Just out of curiosity, the optimal magnification for an APS-C sensor would be 1.6x, correct? I found the below information regarding the adaption of APS-C cameras. Unfortunately this specific configuration with the adapter 7 (T2-T2 DSLR 1.6x #426115) seems not to work on the Axiovert V25 port without some further adaption.

yes 1.6x is the right mag for APS-C.
