Drosophila melanogaster cleared in KOH,whole mount in glicerine.

Wow, great work!

How did you mount such a thick insect, in glycerine, without crashing it, and prevent any subject movement after mounting? Sorry for the newbie question.

I'll just repeat the others.
Fine work, both in terms of photography and as, zzffnn mentioned, in terms of mounting.

I have tried some insect/arthropod mounting and find it hit and miss.

Incredible images! I love the first one. How many stacks was the first image? What were the conditions? Lens etc.Significantly better than any I have produced to date.

Thank you all for your kind comments.

I begin by placing the specimen in KOH/NaOH solution, heat up to ∼90–100 ◦C,5-10min, preferably in a water bath,then washing in pure water.
The most difficult procedure which is a placement and positioning the insect on the slide.
In the center of a microscope slide I put a drop of pure glycerine,transfer the insect by DIY spatula (bend syringe needle) in to glicerine drop.

To positioning the insect under dissecting microscope I use DIY tools ,tattoo needle or cat / dog hair glued to toothpick.

After properly positioning I put very gently the cover slip and sealed with nail varnish

This is not fluorescence,I invert bright field image in Photoshop,and play with the colors.

No doubt those images places you into the world of a fruit fly. The time it takes to do all of this is worth the journey for most of us to view.

I am wondering if you could help us (the newbies) in step-by-step preparation.
I find it hard to keep the sample in place. Or sometimes, the coverslips just crush the fly.
